Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human ZNF217 protein.

Background

Zinc finger protein 217 (ZNF217) is an oncogenic transcription factor that regulates the expression of a wide variety of genes. ZNF217 is a member of the Kruppel-like factor (KLF) family of transcription factors, which consists of zinc finger proteins that bind to CACCC or GT box DNA elements (1). ZNF217 regulates the expression of its target genes through direct DNA binding, and through interaction with transcriptional co-repressors in the CoREST-HDAC complex (2). Although it was initially described as a transcriptional repressor, ZNF217 may also act as a transcriptional activator depending on the target gene (3). Amplification of the ZNF217 gene itself has been observed in a variety of tumors, where it is typically associated with poor prognosis (4-7). Overexpression of ZNF217 protein leads to increased cell proliferation and promotes tumor growth in breast and ovarian cancer (8,9). ZNF217 also interferes with apoptotic signaling and confers resistance to chemotherapeutic drugs in breast cancer (9,10).

Chromatin immunoprecipitations were performed with cross-linked chromatin from MCF7 cells and either ZNF217 (E6X4R) Rabbit mAb or Normal Rabbit IgG #2729 using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR using human CCND1 enhancer primers, and human ZNF10 exon 1 primers. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the total amount of input chromatin, which is equivalent to one.
