    Background

ABT-737 is a BH3 mimetic that is a potent inhibitor of multiple Bcl-2 family members, including Bcl-xL, Bcl-2, and Bcl-w that all have a Ki value of less than or equal to 1 nM (1,3). ABT-737 binds to these anti-apoptotic Bcl-2 family members and inhibits their interaction with pro-apoptotic BH3-domain proteins, leading to Bax-Bak mediated apoptosis. ABT-737 has cytotoxic activity in multiple cancer types, including small-cell lung cancer and lymphoid malignancies (2,3). It has also been shown to inhibit and regress tumor growth in xenograft tumor mouse models (3).
