Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

Phospho-Syntenin-1/MDA9 (Ser60) (F4T2K) Rabbit Monoclonal Antibody recognizes transfected levels of syntenin-1/MDA9 protein only when phosphorylated at Ser60.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic phosphopeptide corresponding to residues surrounding Ser60 of human syntenin-1/MDA9 protein.

Background

Syntenin-1, also known as MDA9 or SDCBP, is a member of the PDZ family of proteins (1), defined by the presence of a PDZ protein interaction domain. Syntenin-1, via its PDZ domain, functions as a scaffold adaptor protein that regulates numerous signal transduction pathways, including FAK, Src, TGF-βR1, EGFR, and IGF1R (2-5). Aberrantly elevated expression of syntenin-1 has been found in various cancers and has been linked to the growth and invasion of tumor cells (5-7). In the central nervous system, syntenin-1 has been shown to mediate neuron dendrite protrusion and spine formation through its interaction with syndecan and Rheb (8). Syntenin-1 is also a key component of the syndecan/syntenin/ALIX pathway, and it has been shown to contribute directly to the formation and internalization of exosomes (9-11).

Syntenin-1/MDA9 has been shown to be phosphorylated at Ser61 by ULK1 in mouse, which is highly conserved with Ser60 in human (12).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
