Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

Rab32 (F6E5T) Rabbit Monoclonal Antibody recognizes endogenous levels of total Rab32 protein. This antibody detects a 55 kDa protein of unknown identity in some cell lines.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Lys208 of mouse Rab32 protein.

Background

Rab32 is a small GTPase best known for its role in regulating membrane trafficking, including endosomal pathway regulation and Golgi apparatus structure and localization (1,2). Rab32 is an essential component of a host defense pathway for defending against intracellular pathogens, helping to link lysosomes and bacteria-containing phagosomes (3,4). Rab32 also recruits IRG1, an enzyme that produces the antimicrobial metabolite itaconate, to pathogen-containing vacuoles (5). Altered Rab32 function is implicated in the pathogenesis of Parkinson's disease. The RAB32 Ser71Arg mutation, which results in increased activation of LRRK2 and reduced colocalization of Rab32 and PINK1, is associated with development of Parkinson's disease (6).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
