Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA, 50% glycerol and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the carboxy terminus of human E2F8 protein.

Background

The E2F family consists of eight transcription factors that regulate gene expression to control cell cycle progression. The atypical E2F family members, E2F-7 and E2F-8, are unique in that they have two distinct DNA-binding domains, but lack the DP-heterodimerization, retinoblastoma-binding, and transcriptional activation domains contained in the canonical E2F family members. E2F-7 and E2F-8 are thought to function primarily as transcriptional repressors (1).
E2F8 is highly expressed in multiple human cancers, including hepatocellular carcinoma, lung, breast, ovarian and cervical cancer, and may serve as a prognostic marker (1-5). In cancer cell lines, E2F8 expression enhances proliferation, migration, invasion, and the expression of epithelial mesenchymal transition (EMT) markers (2,3).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Immunoprecipitation of E2F8 protein from HT-29 cell extracts. Cells were treated with Aphidicolin #32774 (10 μg/mL, 24 hr). Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is E2F8 (E8I3L) Rabbit mAb. Western blot analysis was performed using a polyclonal anti-E2F8 antibody. Anti-rabbit IgG, HRP-linked Antibody #7074 was used as the secondary antibody.
