Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Phe164 of human FIP1L1. Antibodies are purified by protein A and peptide affinity chromatography.

Background

FIP1L1 is the mammalian homolog of yeast Fip1p (factor interacting with poly(A) polymerase (PAP)). It is a subunit of cleavage and polyadenylation specificity factor (CPSF) that binds to PAP and to U-rich sequences on the pre-mRNA, stimulating polyadenylation of pre-mRNA (1). FIP1L1 has several splicing variants (1). FIP1L1 is fused to platelet-derived growth factor receptor α gene (PDGFRα) by an interstitial deletion on chromosome 4q12 in some patients with idiopathic hypereosinophilic syndrome (HES) and chronic eosinophilic leukemia (CEL). Τhe resulting FIP1L1-PDGFRα fusion protein has constitutively elevated tyrosine kinase activity (2).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
