Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the amino terminus of human PSPC1 protein.
Background
PSPC1 (paraspeckle component 1) was one of the first proteins found to be enriched in subnuclear bodies that are adjacent to but distinct from nuclear speckles, and subsequently named paraspeckles (1). PSPC1 belongs to the Drosophila melanogaster behavior protein family comprised of human splicing (DBHS) proteins, including SFPQ (splicing factor proline-glutamine rich ) and NONO (non-POU domain-containing octamer-binding protein) which are also enriched in paraspeckles (2). DBHS proteins are a unique family of RNA-binding proteins that recruit various proteins and nucleic acids involved in transcriptional activation and repression, transcript localization, splicing, and DNA damage repair (3). Important to disease, DBHS proteins bind the long noncoding RNA NEAT1 which is found to be enriched in paraspeckles. Aberrant accumulation of NEAT1 transcripts in paraspeckles has been identified in multiple tumor types, leading to a proposal that the long coding RNA, via the process of sponging, disrupts the ratio of miRNAs to target proteins, resulting in altered protein expression profiles that promote cancer (4).
Species Reactivity
Species reactivity is determined by testing in at least one approved application (e.g., western blot).
Western Blot Buffer
IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
