Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

RNF128 Antibody recognizes endogenous levels of total RNF128 protein. This antibody detects a 40 kDa band of unknown origin in some cell lines.

Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human RNF128 protein. Antibodies are purified by peptide affinity chromatography.

Background

RNF128, also known as gene related to anergy in lymphocytes (GRAIL), is a RING finger protein family E3 ubiquitin ligase that participates in T cell anergy induction (1,2). RNF128 is a type I transmembrane protein, made up of a zinc RING finger, coiled-coil, and protease-associated domain (1,2). Expression of RNF128/GRAIL has been shown to inhibit cytokine transcription and CD4 T cell proliferation, as well as regulate naïve T cell tolerance induction and Treg cell function (2-4).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
