Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA, 50% glycerol and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

SLFN11 (D8W1B) Rabbit mAb recognizes endogenous levels of total SLFN11 protein. Non-specific staining was observed in skeletal muscle.

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to human SLFN11 protein.

Background

SLFN11 is a nuclear protein that belongs to the Schlafen (SLFN) family of genes involved in cell cycle regulation and growth inhibition (1,2). Expression of SLFN11 predicts sensitivity of cancer cell lines to DNA-damaging agents (1,3). Evidence suggests that in the presence of DNA-targeted therapies, SLFN11 is recruited to stressed replication forks where it blocks replication, leading to cell death (4). SLFN11 is being explored as a predictive biomarker for response to DNA-targeted therapies (5).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
