Specificity / Sensitivity

TRIM9 Antibody recognizes endogenous levels of total TRIM9 protein. TRIM9 Antibody is predicted to cross-react with all three TRIM9 isoforms.

Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val217 of human TRIM9 protein. Antibodies are purified by peptide affinity chromatography.

Background

Tripartite motif-containing protein 9 (TRIM9) is a brain-enriched E3 ubiquitin-protein ligase. TRIM family proteins contain a zinc-binding domain, followed by RING finger, B-box, and coiled-coil domains (1). TRIM proteins are implicated in several biological processes, including cell development and growth, but their molecular mechanism of action remains unclear. Interestingly, TRIM9 is reported to function as an E3 ubiquitin ligase (2). Ubiquitination is the post-translational modification of protein substrates by ubiquitin, a 76-amino acid polypeptide. This process is important for cellular protein turnover, and alterations in this process are linked to human diseases, including neurodegenerative diseases (3). Protein ubiquitination is mediated by a trio of ubiquitination enzymes, including E3 ubiquitin ligases. As a brain-enriched E3 ubiquitin ligase, TRIM9 may play a role in maintaining proper cellular proteostasis in the brain (2). Alterations in TRIM9 expression may contribute to human neurodegenerative diseases, including Parkinson's disease, or injury (2,4).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
