Specificity / Sensitivity

FBP2/FBPase 2 (E9M5T) Rabbit mAb recognizes endogenous levels of total FBP2/FBPase 2 protein. This antibody does not cross-react with FBP1/FBPase 1 protein. This antibody detects an 80 kDa nonspecific band.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the amino terminus of human FBP2/FBPase 2 protein.

Background

Fructose-1,6-bisphosphatase 2 (FBP2 or FBPase 2) is a rate limiting enzyme in gluconeogenesis. It catalyzes the conversion of fructose-1,6-bisphosphate to fructose-6-phosphate (1,2). FBP2/FBPase 2 expression is inhibited in various soft tissue sarcoma subtypes. Ectopic expression of FBP2/FBPase 2 represses soft tissue sarcoma cell proliferation and tumor growth. Studies show that cytosolic FBP2/FBPase 2 inhibits glycolysis through its catalytic activity, suppressing sarcoma cell proliferation. In addition, nuclear FBP2/FBPase 2 inhibits mitochondrial biogenesis and oxidative phosphorylation independent of its catalytic activity, also contributing to the suppression of sarcoma cell proliferation (2).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
