Target Information

a constitutively active proline-directed protein kinase. Involved in energy metabolism, neuronal cell development, and body pattern formation. Acts as a negative regulator in the hormonal control of glucose homeostasis, Wnt signaling, and regulation of transcription factors and microtubules. Regulates protein synthesis by controlling the activity of EIF2B. In Wnt signaling, GSK3B phosphorylates CTNNB1 leading to its degradation. Phosphorylates and inactivates APC, AXIN1, CRMP2, JUN, NFATC, MAPT and MACF1. Is necessary for the establishment of neuronal polarity and axon outgrowth. Regulates the circadian clock via phosphorylation of major clock components. Acts as a regulator of autophagy by activating TIP60, thus promoting the acetylation of key autophagy regulators. Expressed in testis, thymus, prostate and ovary and weakly expressed in lung, brain and kidney. Colocalizes with EIF2AK2/PKR and TAU in the Alzheimer disease (AD) brain. Most of its substrates require priming by phosphorylation of a residue C-terminal to the targeted serine or threonine. Belongs to the protein kinase superfamily, CMGC Ser/Thr kinase family, GSK-3 subfamily. 2 alternatively spliced human isoforms have been reported.
