    This Cell Signaling Technology® antibody is conjugated to the carbohydrate groups of horseradish peroxidase (HRP) via its amine groups. The HRP conjugated antibody is expected to exhibit the same species cross-reactivity as the unconjugated 3X DYKDDDDK Tag (E7C5T) Rabbit mAb #87537.

    Storage

    Supplied in 136 mM NaCl, 2.6 mM KCl, 12 mM sodium phosphate (pH 7.4) dibasic, 2 mg/mL BSA, and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    3X DYKDDDDK Tag (E7C5T) Rabbit mAb (HRP Conjugate) detects exogenously expressed proteins containing the 3X DYKDDDDK epitope tag. This antibody recognizes the 3X DYKDDDDK tag fused to either the amino or carboxy terminus of targeted proteins in transfected cells, but does not detect DYKDDDDK tag. 3X DYKDDDDK Tag (E7C5T) Rabbit mAb (HRP Conjugate) detects exogenously expressed 3X DYKDDDDK-tagged proteins in cells expressed under a CMV promoter. Expression under other promoters has not been evaluated.

    Monoclonal antibody is produced by immunizing animals with a synthetic peptide containing the 3X DYKDDDDK epitope (DYKDHDGDYKDHDIDYKDDDDK).

    Background

    Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein’s biochemical properties.

    The DYKDDDDK peptide has been used extensively as a general epitope tag in expression vectors. This peptide can be expressed and detected with the protein of interest as an amino-terminal or carboxy-terminal fusion (1).
