    Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    Phospho-PDK1 (Tyr373/376) Antibody detects endogenous levels of PDK1 when only phosphorylated at tyrosine 373/376. This antibody may cross-react with some other proteins. CST recommends an immunoprecipitation of PDK1 prior to Western immunoblotting.

    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic phosphopeptide corresponding to residues surrounding Tyr373/376 of human PDK1. Antibodies are purified by protein A and peptide affinity chromatography.

    Background

    Phosphoinositide-dependent protein kinase 1 (PDK1) plays a central role in many signal transduction pathways (1,2) including the activation of Akt and the PKC isoenzymes p70 S6 kinase and RSK (3). Through its effects on these kinases, PDK1 is involved in the regulation of a wide variety of processes, including cell proliferation, differentiation and apoptosis.
    PDK1 becomes phosphorylated at Tyr373/376 and translocates to the plasma membrane in response to pervanadate or insulin treatment. Mutational analysis of these sites indicates that Tyr373/376 is important for PDK1 catalytic activity (4).
