    Product Description

    PathScan® Total PCNA Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of PCNA. A PCNA Rabbit mAb has been coated onto the microwells. After incubation with cell lysates, PCNA protein is captured by the coated antibody. Following extensive washing, a PCNA Mouse Detection mAb is added to detect the captured PCNA protein. A HRP-linked, anti-mouse antibody is then used to recognize the bound detection antibody. HRP substrate, TMB, is added to develop color. The magnitude of the absorbance for this developed color is proportional to the quantity of total PCNA.
    Antibodies in kit are custom formulations specific to kit.

    Protocol

    Specificity / Sensitivity

    PathScan® Total PCNA Sandwich ELISA Kit detects endogenous levels of PCNA protein as shown in Figure 1. This kit sensitivity is shown in Figure 2. This kit detects proteins from the indicated species, as determined through in-house testing, but may also detect homologous proteins from other species.

    Background

    Proliferating cell nuclear antigen (PCNA) is a member of the DNA sliding clamp family of proteins that assist in DNA replication (1). Crystal structure data suggests that a PCNA homotrimer ring can encircle and slide along the DNA double helix (2). Multiple proteins involved in DNA replication, DNA repair, and cell cycle control bind to PCNA rather than directly associating with DNA, thus facilitating fast processing of DNA (reviewed in 3). PCNA protein expression is a well-accepted marker of proliferation.
