    Prior to use, thaw the recombinant protein and briefly spin down to collect all liquid at the bottom of the tube. Dilute according to the following instructions depending on the ELISA kit type. An example sensitivity curve is shown in Figure 2.

    PathScan® ELISA Kits:

    Dilute with ELISA Sample Diluent to achieve a 1X protein concentration.

    PathScan® RP ELISA Kits:

    Dilute with 1X Cell Lysis Buffer #9803 to a 2X protein concentration to achieve a final 1X protein concentration upon addition of the detection antibody.

    FastScan™ ELISA Kits:

    Dilute with 1X FastScan™ ELISA Cell Extraction Buffer to a 2X protein concentration to achieve a final 1X protein concentration upon addition of the antibody cocktail..

    Human CD3ζ (R52-R164)-Whitlow Recombinant Protein Quantitative Control is derived from a recombinant expression construct with CD3ζ (R52-R164) and Whitlow. The protein can be used with PathScan® RP Whitlow/218 Linker/CD3ζ CAR Sandwich ELISA Kit #72378 and Whitlow/218 Linker/CD3ζ CAR Matched Antibody Pair #97322 as a quantitative control. Suitable for use in other Whitlow/218 Linker/CD3ζ assays when a control is needed.

    Background

    The Whitlow/218 peptide is a commonly used linker that connects the variable heavy (VH) domain and variable light (VL) domain of single-chain variable fragments (scFvs) and chimeric antigen receptors (CARs) that utilize an extracellular domain scFv for target antigen recognition. The Whitlow/218 linker affords the scFv with enhanced resistance to proteolysis and aggregation (1).
