    Background

    The adaptor-protein 2 (AP-2) coat assembly protein complex is an important component of clathrin-coated pits involved in receptor-mediated endocytosis at the plasma membrane (1-3). Each AP-2 heterotetramer is composed of α, β, μ, and σ protein subunits. AP-2 is involved in clathrin-dependent endocytosis in which cargo proteins are incorporated into clathrin-coated vesicles destined for incorporation into the endosomal compartment. AP-2 complex subunit alpha-2 (AP2A2) is one of two large alpha-type adaptins that exist within the AP-2 complex. Research studies have shown that AP2A2 works in concert with AP-2 sigma subunits to facilitate the recognition of [ED]-X-X-X-L-[LI] endocytic motifs embedded within the cytoplasmic tails of membrane cargo proteins destined for the endosomal compartment (4).
