    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val188 of human dUTPase/DUT protein.

    Background

    dUTP pyrophosphatase (dUTPase), also known as deoxyuridine triphosphatase (DUT), catalyzes the conversion of dUTP to dUMP and pyrophosphate (1,2). Studies show that fumarate, an intermediate in the TCA/Krebs cycle, functions as a signaling metabolite to regulate mitochondrial biogenesis. Fumarate binds to the mitochondrial enzyme, malic enzyme 2 (ME2), and stimulates the formation of ME2 dimers. ME2 dimers, in turn, activate the enzymatic activity of dUTPase/DUT, promoting thymidine biosynthesis and increasing mitochondrial DNA replication (3).
