    Background

    ETS-related transcription factor Elf-4 (ELF4), also known as myeloid Elf-1-like factor (MEF), is a member of the ETS-family of transcription factors, all containing a well-conserved ETS DNA-binding domain that recognizes a core consensus sequence defined in vitro as 5-’GGAA’ (1,2). In addition to being a transcriptional activator, ELF4 also plays many roles within the immune system, such as regulating natural killer (NK) cell development and perforin expression, CD8+T cell proliferation, anti-viral immunity through type I interferon, and TH17 cell differentiation (1-6). Lack of ELF4 in humans has been associated with deficiency in ELF4, X-linked (DEX), a monogenic autoinflammatory disease (mAID) (5,6). In mouse models, Elf4 KO macrophages failed to sustain the transcription of key anti-inflammatory genes and instead upregulated proinflammatory genes (5).
