    Background

    Ataxia telangiectasia and Rad3-related (ATR) is an essential kinase with functions in DNA damage and the replication stress response, as well as in maintaining chromosome stability during mitosis (1,2). ATR is activated in mammalian cells by TopBP1 (3) or Ewing's tumor-associated antigen 1 (ETAA1) (4). ETAA1 binds to RPA-coated single-stranded DNA at stalled replication forks, activating ATR signaling independently of TopBP1 in response to replication stress, protecting replication fork integrity (4-6). ATR activation by ETAA1 is required for T cell clonal expansion in mice (7). Quantitative phosphoproteomics demonstrates a dominant role for ETAA1 in mitotic ATR activation (8).
