    Protocol

    Specificity / Sensitivity

HAND1 (E7C4L) Rabbit mAb recognizes endogenous levels of total HAND1 protein. This antibody may detect nonspecific bands at 140 and 200 kDa.

    Species Reactivity:

    Human

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly79 of human HAND1 protein.

    Background

HAND1 is a basic helix-loop-helix (bHLH) transcription factor (TF) that is capable of forming heterodimers with other binding partners, functioning as a transcriptional activator or a repressor depending on the underlying sequence and dimerization partner (1,2). HAND1 is a key regulator of trophoblast giant cell differentiation and cardiac morphogenesis (3). Moreover, several studies have demonstrated that this TF plays an essential role in cell proliferation and carcinogenesis. Namely, HAND1 has been reported to be downregulated in several cancers, including pancreatic, colorectal, and small cell lung cancers (4-7). Furthermore, HAND1 gene expression is negatively regulated by the high mobility group A1 (HMGA1) protein in human thyroid carcinomas (8) and its expression was shown to be necessary in sustaining gastrointestinal stromal tumor (GIST) proliferation (9).
