Product Usage Information

This product is the carrier free version of product #95153. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

Storage

Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the amino terminus of human integrin β6 protein.

Background

Integrins are α/β heterodimeric cell surface receptors that play a pivotal role in cell adhesion and migration, as well as in growth and survival (1,2). The integrin family contains at least 18 α and 8 β subunits that form 24 known integrins with distinct tissue distribution and overlapping ligand specificities (3). Integrins not only transmit signals to cells in response to the extracellular environment (outside-in signaling), but also sense intracellular cues to alter their interaction with extracellular environment (inside-out signaling) (1,2).

Integrin β6 pairs with integrin α5. Expression of this dimer occurs exclusively on injured epithelium cells, and is present only at very low levels on normal tissue. It is expressed during development, but expression becomes repressed in adulthood (3). It is involved in wound healing, fibrosis, pulmonary fibrosis in particular, and tumorigenesis (4-6). When the dimer is unregulated in the membrane in response to injury, the integrin β6 subunit binds directly to an RGD motif present in fibronectin and TGF-β, both present in the extracellular matrix. Integrin β6 cleaves and binds the LAP fragment of latent TGF-β, thereby releasing active TGF-β into the ECM to activate myofibroblasts which consequently mediate ECM remodeling (3-6).
