    Background

    Lysophospholipase-like 1 (LYPLAL1) is a serine hydrolase that has been linked to various metabolic disorders, including insulin resistance and non-alcoholic fatty liver disease (1,2). Although it is evolutionarily conserved, knockout studies in mice confirm that LYPLAL1 is not essential for normal metabolic function (3). LYPLAL1 has also been shown to depalmitoylate cyclic GMP-AMP synthase (cGAS), inhibiting cGAS enzymatic activity and downstream signaling (4). Inhibition of LYPLAL1 results in enhanced cGAS-mediated innate immune signaling, suggesting that this treatment could be used to bolster anti-tumor immunotherapies such as PD-1 blockade (4).
