    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val331 of human NADK2 protein.

    Background

    NAD kinase 2 (NADK2) is a mitochondrial enzyme that catalyzes the phosphorylation of NAD+ to NADP+. This enzyme uses ATP or inorganic polyphosphate as a phosphoryl donor. Besides NAD+, NADK2 can also use NADH as a substrate (1). NADK2 is essential for NADP+ and NADPH generation in mitochondria. Studies show that tricarboxylic acid cycle activity, mitochondrial folate pathway activity, and oxidative stress in mitochondria are not affected when the NADK2 gene is deleted. However, NADK2 deletion leads to diminished NADP+ and NADPH in mitochondria and negatively impacts the reduction of glutamate to pyrroline-5-carboxylate needed for proline biosynthesis. As a result, NADK2 deletion slows the proliferation of cells cultured in minimal medium (2).
