Background
RNF114 is involved in various biological processes, including embryonic development (3,4), cell proliferation (5), genomic integrity (6,7), the immune response (8), and osteoclastogenesis (9). It functions by adding ubiquitin chains to substrates such as A20, TRAF6, PARP1, and CDKN1A (p21), thereby modulating the NF-κB, DNA damage response, and cell cycle pathways. Furthermore, RNF114 facilitates viral infections by targeting the host mitochondrial antiviral signaling protein (MAVS) for degradation (10,11).
Aberrantly high expression of RNF114 is associated with breast, gastric, and colon cancers. In these contexts, RNF114 promotes cancer cell proliferation, invasion, and metastasis by interacting with or degrading cell-type-specific signaling molecules, including EWSR1, EGR1, and junction plakoglobin (JUP) (12-14).
