    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ala756 of human SAFB2 protein.

    Background

    Scaffold attachment factor B1 and B2 (SAFB1/B2) are homologs that bind to scaffold or matrix attachment region DNA elements (S/MAR DNA) (1,2). SAFB1 and SAFB2 bind to estrogen receptor to inhibit its activity, an action enhanced by tamoxifen treatment and opposed by histone deacetylase activity (2-4). SAFB1 and SAFB2 contain an RNA recognition motif and can interact with various hnRNP complexes and serine/arginine-rich splicing factors to influence pre-mRNA splicing (5-7). SAFB1 is also capable of helping in the DNA damage response by allowing for yH2AX spreading (8). Knockdown of SAFB1 results in increased SAFB2 expression, and knockdown of SAFB2 results in increased SAFB1 expression (9). Despite their high homology, SAFB1 and SAFB2 have some non-redundant functions. SAFB1 knockout mice have a high degree of growth and fertility issues, while SAFB2 knockout mice have a normal phenotype but aberrations in Sertoli cells (10). SAFB proteins are expressed lowly in breast cancers with poor prognosis (11).
