    Background

    Semaphorin-7A is a GPI-anchored membrane protein that plays important roles in neuronal development and immune responses via integrin-mediated signaling. It promotes axon growth by activating the mitogen-activated protein kinase (MAPK) pathway through β1 integrin receptors. When expressed on activated T cells, it promotes the production of proinflammatory cytokines by macrophages through the α1β1 integrin receptor (1). Semaphorin-7A may also play a role in resolving inflammation through its regulation of macrophage chemotaxis and chemokinesis, and by promoting macrophage differentiation and polarization toward an anti-inflammatory phenotype (2). Neuroinflammation is a detrimental component of neurodegenerative diseases, such as Parkinson’s disease (PD) and multiple sclerosis (MS). The role that semaphorin-7A plays in mediating inflammatory responses in microglia in the brain suggests that it may be a therapeutic target for these diseases. In a cell model of neuroinflammation, semaphorin-7A was shown to promote cell viability and survival through its mediation of MAPK and peroxisome proliferator-activated receptor γ (PPARγ) pathways (3).
