Product Usage Information

This product is the carrier free version of product #20407. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

Storage

Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Leu264 of human SerpinB1 protein.

Background

Serpin family B member 1 (SerpinB1), also known as leukocyte elastase inhibitor or neutrophil elastase inhibitor, regulates inflammation and the innate immune response. SerpinB1 blocks the activity of caspase-1, caspase-4, and caspase-5; the protein does so via suppression of caspase recruitment domain oligomerization and enzymatic activation (1). SerpinB1 is expressed in neutrophils and monocytes, and the protein resides in the cytoplasm due to its lack of a secretory signal sequence (2,3). There, SerpinB1 protects cells from serine proteases that may leak from granules into the cytoplasm, including Cathepsin G, neutrophil elastase, proteinase-3, and Granzyme H (4,5). Despite its lack of a secretory sequence, SerpinB1 was shown to be released from keratinocytes in a caspase-1-dependent manner to upregulate proliferation of pancreatic β cells (6).
