    Background

    SHC-transforming protein 1 (SHC1) is an adaptor protein to targets of activated tyrosine kinases. SHC1 is ubiquitously expressed and the gene is expressed in 2 transcripts, resulting in 3 isoforms with distinct functions and subcellular locations (1). The phosphorylated p46 and p52 isoforms couple activated receptor tyrosine kinases to Ras via the recruitment of the GRB2/SOS complex. The third isoform, p66, acts as a downstream target of the tumor suppressor p53 and is thereby implicated in oxidative stress response, tumor progression, and cancer cell proliferation (2,3). SHC1 has been identified to have prognostic and diagnostic value in multiple cancers, making it a potential biomarker (4).
