    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Lys346 of human Shroom3 protein.

    Background

    Shroom protein family members (Shroom1-4) regulate early developmental cell shape and are characterized by variable N-terminal PDZ domains, centrally positioned ASD1 domains, and C-terminal ASD2 domains (1). Shroom3 (formerly Shroom) contains all thee domains, with ASD1 serving as an F-actin binding domain, while ASD2 is required for apical membrane constriction (2). The Shroom3 ASD2 domain directly binds Rho-associated kinases ROCK1 and ROCK2 (3), driving myosin II and myosin light chain activity at apical cell junctions (4). Shroom3 expression in polarized epithelial cells is necessary and sufficient to drive neural tube closure in developing vertebrates (5). In developing mouse eye, Shroom3 is recruited by catenin δ-1 (p120 catenin) to epithelial adherens junctions, where both proteins are required for lens pit invagination (6). Shroom3 is required for proper development of glomerular and renal tube structure (7,8), and knockdown of Shroom3 leads to disruption of podocyte lamellipodia formation (9).
