    Background

    β-II spectrin (SPTBN1), the most common isoform of non-erythrocyte spectrin, is a cytoskeleton protein in many cell types (1). SPTBN1 is a large actin cross-linking and scaffold protein that links the cell membrane and cytoskeleton. It is crucial for not only cell shape and organization but also a variety of cellular functions, such as cell adhesion, spreading, cell cycle regulation, and apoptosis (1,2).

    SPTBN1 is involved in cell growth and development (particularly in brain) via the TGF-β/Smad signaling pathway (3,4). SPTBN1 knockout mice show defects in cortical organization, developmental delay, and behavioral deficiencies (3), and variants in human SPTBN1 have been strongly implicated in intellectual disabilities and autism (4). Aberrant expression of SPTBN1 in various cell and tissue types has been shown in cancer development and progression, particularly ovarian, breast, and liver cancers (5-8). SPTBN1 plays an anticancer role in many tumor types. It assists in chemotherapeutic resistance and decreases the migration ability of tumor cells (5,6), highlighting its role in metastatic tumor progression and its potential as a pharmaceutical target (7-9). Recent work has shown that SPTBN1 is important for controlling CAR-T cell migration targeting tumors (10).
