    Background

    Serine palmitoyltransferase 2 (SPTLC2) is a component of the serine palmitoyltransferase (SPT) multisubunit enzyme, the principal enzyme facilitating the initial, rate-limiting step in de novo sphingolipid biosynthesis (1,3-5). This protein complex consists of an SPTLC1/SPTLC2 or SPTLC1/SPTLC3 heterodimer as the catalytic core (1-3). Sphingolipid biosynthesis generates long-chain based compounds by condensing serine with activated acyl-CoA to form ketodihydrosphingosine (3,4). SPTLC2 mutations have been linked to hereditary sensory and autonomic neuropathy type I (HSAN1), frontotemporal dementia (FTD), and early-onset amyotrophic lateral sclerosis (ALS) (2,5).
