    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding cysteine 57 of human TRAF1. Antibodies are purified by protein A and peptide affinity chromatography.

    Background

    TRAFs (TNF receptor-associated factors) are a family of multifunctional adaptor proteins that bind to surface receptors and recruit additional proteins to form multiprotein signaling complexes capable of promoting cellular responses (1-3). Members of the TRAF family share a common carboxy-terminal "TRAF domain", which mediates interactions with associated proteins; many also contain amino-terminal Zinc/RING finger motifs. The first TRAFs identified, TRAF1 and TRAF2, were found by virtue of their interactions with the cytoplasmic domain of TNF-receptor 2 (TNFRII) (4). The six known TRAFs (TRAF1-6) act as adaptor proteins for a wide range of cell surface receptors and participate in the regulation of cell survival, proliferation, differentiation, and stress responses.
    TRAF1 is unique amongst the TRAF family in that it does not contain an amino-terminal RING finger (4). TRAF1 has a limited tissue distribution and may be induced by NF-kappaB and inactivated by lymphocytes (4-6). TRAF1 works in conjunction with TRAF2 to suppress caspase activation and TNF-induced apoptosis. It has been shown to associate with a number of cytoplasmic proteins, including TANK/I-TRAF, TRIP, A20, RIP, RIP2, FLIP and c-IAP1/2. During apoptosis, TRAF1 may be cleaved by caspases at Asp163 which can act to inhibit NF-kappaB activation (7,8).
