    Protocol

    Specificity / Sensitivity

TRAP/ACP5 (F7J8N) Rabbit Monoclonal Antibody recognizes endogenous levels of total TRAP/ACP5 protein. This antibody detects both the proenzyme (TRAP 5a) and the mature enzyme (TRAP 5b).

    Species Reactivity:

    Human, Mouse

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Asn131 of human TRAP/ACP5 protein.

    Background

Tartrate-resistant acid phosphatase type 5 (TRAP/ACP5) is a metalloenzyme synthesized as a roughly 35 kDa proenzyme, TRAP 5a, that is later processed into a 17 kDa mature or active form, TRAP 5b (1-3). TRAP 5b serum levels correlate to osteoclast numbers, though TRAP/ACP5 is also expressed in other cell types, including macrophages and dendritic cells (4,5). TRAP/ACP5 plays an essential role in regulating bone resorption by dephosphorylating osteopontin to promote osteoclast migration (6). In macrophages, TRAP/ACP5 helps regulate the secretion of inflammatory cytokines (7). In dendritic cells, TRAP/ACP5 is important for maturation (8). TRAP/ACP5 can also promote breast cancer metastasis and pulmonary fibrosis through activation of Smad2/3, TGFβR2, and β-catenin signaling (9,10).
