    Background

    Tetraspanin 8 (TSPAN8) belongs to the tetraspanin family, which is characterized by four transmembrane domains, one short extracellular domain (ECL1), and one long extracellular domain (ECL2) (1,2). Tetraspanins interact with a variety of cell surface proteins and intracellular signaling molecules in specialized tetraspanin-enriched microdomains (TEMs) where they mediate a range of processes, including adhesion, motility, membrane organization, and signal transduction (2,3). TSPAN8 is overexpressed in numerous epithelial cancers, where it promotes cell invasion and metastasis and is associated with poor clinical outcomes (4-6). TSPAN8 also contributes to stemness in cancer stem cells (CSCs) through activation of β-catenin and Sonic hedgehog (Shh) signaling pathways (7,8). CSCs further secrete TSPAN8 on the surface of exosomes, enhancing stemness in the surrounding healthy tissue (9). To this end, TSPAN8 may serve as a useful blood biomarker for the early detection of cancer cells or CSCs (10,11). Therapeutics targeting TSPAN8 have also shown initial success in treating cancer, including monoclonal antibodies, bispecific antibodies, and chimeric antigen receptor (CAR) T cells (12-14).
