    Background

    UDP-N-acetylglucosamine pyrophosphorylase 1 (UAP1) is a metabolic enzyme that plays a key role in the hexosamine biosynthesis pathway (HBP). UAP1 catalyzes the final step in the pathway by converting uridine triphosphate (UTP) and GlcNAc-1-phosphate to UDP-GlcNAc, a nucleotide sugar (1). UDP-GlcNAc is further utilized as a substrate for multiple glycosylation reactions, including O-GlcNAcylation of target proteins (2). HBP dysfunction has been linked to various cancers, potentially due to elevated O-GlcNAcylation levels affecting signaling pathways (3). UAP1 expression levels are particularly elevated in prostate cancer, and knockdown of UAP1 has been found to reduce cell growth and viability in prostate cancer cells (4-6). Altered UAP1 expression levels have also been linked to other cancers, such as osteosarcoma and breast cancer (7,8).
