How do I lyse my cells for the Senescence β-Galactosidase Activity Assay Kit (Fluorescence, Plate-Based) #23833?

To lyse cells for the Senescence β-Galactosidase Activity Assay Kit (Fluorescence, Plate-Based) #23833:

1.  Aspirate cell growth media.
2.  Rinse cells with 100uL of ice cold 1XPBS.
3.  Add 100uL of ice cold Senescence Lysis Buffer to each well.
4.  Place 96 well plate(s) on ice and incubate for 5 min.
5.  Aspirate lysis buffer up and down several times to break up cells.
6.  Centrifuge 96 well plate(s) 5 min at 1200 rpm to pull down any cellular debris.
7.  Remove cell lysate supernatant and add desired amount to the assay.
 
These steps can also be adapted for larger assay plates by adjusting the volume of PBS and lysis buffer accordingly.
