How should I resuspend and aliquot the PTMScan® LysC Protease #84748?

To resuspend the PTMScan® LysC Protease #84748, we recommend allowing the vial to equilibrate at room temperature for a few minutes after removing from the -20C freezer. Next, spin the vial at 1,000 x g for 1 min prior to opening the cap. Dispense the HEPES buffer with non-autoclaved pipette tips, vortex to make sure the powder is resuspended while avoiding excess bubbles, centrifuge the vial at 1,000 x g for 1 min, and aliquot into non-autoclaved microcentrifuge tubes. 

For PTMScan kits that recommend 10-20 mg of sample peptide per assay, such as PTMScan Phospho-Akt Substrate Motif mAb 2 (RXRXXS*/T*) Kit #5563, you will need 40-80 µg of LysC per assay. Thus, the 1 mg LysC vial should be dissolved in 0.2 mL of HEPES and split across twelve 16 µL aliquots (for 80 µg aliquots) or twenty-four 8 µL aliquots (for 40 µg aliquots).
