Will your Phospho-Smad1/5 (Ser463/465) (41D10) Rabbit mAb #9516 cross-react with Smad9?
The amino acid region surrounding the Ser463/465 phospho sites is well conserved in Smad1, 5, and 9. An alignment using the human sequences found in UniProt shows only one residue difference in this region. While we have performed testing to demonstrate that our Phospho-Smad1 (Ser463/465)/ Smad5 (Ser463/465)/ Smad9 (Ser465/467) (D5B10) Rabbit mAb #13820 binds phospho-Smad9, we have not performed similar testing with our Phospho-Smad1/5 (Ser463/465) (41D10) Rabbit mAb #9516. Based on sequence conservation we would predict that #9516 may cross-react with phospho-Smad9, as we indicate in the Specificity/Sensitivity statement on our datasheet and product webpage. However, until we perform the necessary experiments we cannot guarantee phospho-Smad9 recognition using #9516.
