    This Cell Signaling Technology® antibody is the trFluorTM Europium Cryptate version of the unconjugated CRBN (D8H3S) Rabbit mAb #71810 and is expected to exhibit the same species cross-reactivity. Time-resolved (TR) fluorescence resonance energy transfer (FRET), or TR-FRET, data were generated using the conjugated antibody.

    Product Usage Information

    trFluorTM Europium Cryptate conjugated antibodies are ideal for protein-protein interaction assays, pair-based immunoassay technologies, or high-throughput screening where greater sensitivity and quantitative detection is needed. The long fluorescent decay time of Europium Cryptate, enhanced with buffer containing Potassium fluoride, results in improved signal to background over fluorescent dye conjugates. Methods utilizing Europium Cryptate conjugated antibodies include, but are not limited to, TR-FRET. In TR-FRET assays, Europium Cryptate-labeled donor antibodies are paired with red fluorophore-labeled acceptor antibodies, such as Alexa Fluor® 647 or APC.


    Please contact us if you require an acceptor antibody clone conjugated to a red dye at a custom concentration, a carrier-free formulation, or a more customized packaging solution. Optimal dilutions and or working concentrations should be determined by the end user.

    Storage

    Supplied in PBS (pH 7.2), less than 0.1% sodium azide, and 2 mg/mL BSA. Store at 4°C. Do not aliquot the antibody. Protect from light. Do not freeze.

    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro44 of human CRBN protein.

    Background

    CRBN/Cereblon, a substrate receptor for the CRL4 E3 ubiquitin ligase, was originally identified for its role in autosomal recessive nonsyndromic mental retardation (1). CRBN’s role in the regulation of the AMPK pathway has been linked to various human diseases, including cardiovascular disease and obesity, and its involvement in proteasomal degradation may be important in neuropsychiatric diseases (2). CRBN has also been shown to enhance the effects of immunomodulatory imide drugs (IMiDs) in multiple myeloma, and may be a predictive marker for therapy (2-4).
