    Background

    3 beta-hydroxysteroid dehydrogenase/Delta 5-->4-isomerase type 1 (HSD3B1), also known as 3β-hydroxysteroid dehydrogenase-1 (3βHSD1), is critical for steroid hormone biosynthesis (1,2). This enzyme catalyzes the rate-limiting step of extragonadal androgen synthesis. Studies show that glucosamine released from cancer-associated fibroblasts (CAFs) enhances O-GlcNAcylation in prostate cancer cells, activating HSD3B1/3βHSD1 expression mediated by the transcription factor Elk1. This increases de novo intratumoral androgen biosynthesis and leads to castration-resistant prostate cancer (CRPC) (2). In addition, the adrenal-permissive HSD3B1 (1245C) allele, which upregulates intratumoral androgen biosynthesis, augments resistance to radiotherapy in prostate cancer (3).
