    Background

    Hyaluronan and proteoglycan link protein 1 (HAPLN1) is a critical component of the extracellular matrix (ECM), functioning by cross-linking hyaluronic acid (HA) and various proteoglycans (1). The protein promotes the formation of aggregate structural complexes within the ECM, which are essential for cartilage formation and neocortex folding during development (2,3). Age dependent loss of HAPLN1 increases lymphatic and vascular permeability (4,5). Additionally, HAPLN1 protects cell-surface TGF-beta receptors, thereby maintaining TGF-beta signaling, contributing to the rejuvenation of aged skin and the mitigation of pulmonary emphysema (6,7). While increased expression of HAPLN1 is linked to cancer invasion, metastasis (8-10), therapeutic drug resistance (11), and inflammatory arthritis (12,13), knockdown of HAPLN1 has been shown to inhibit the development of heart failure by activating the PKA signaling pathway (14,15).
