    Background

    Calcium-binding and coiled-coil domain-containing protein 1 (CALCOCO1), also known as CoCoA, is a versatile adaptor protein that plays a critical role in cellular homeostasis. It was initially characterized as a coactivator for various transcription factors, including androgen receptor and β-catenin, and functions by recruiting histone acetyltransferases, such as p300 and CBP, to promoters of genes involved in cell growth and development (1-3).

    More recently, mass spectrometry proteomic profiling aimed at identifying proteins in the mTOR-autophagy axis revealed a role for CALCOCO1 in selective autophagy (4). Notably, CALCOCO1 acts as a soluble reticulophagy receptor for the selective degradation of tubular endoplasmic reticulum (ER) in response to proteotoxic and nutrient stress. CALCOCO1-mediated ER-phagy requires interaction with VAMP-associated proteins on the ER membrane via its C-terminal FFAT-like motif, followed by binding to ATG8/LC3 proteins on the autophagosome via its LC3-interacting region (LIR) motif (5). Additionally, CALCOCO1 binds the Golgi-resident palmitoyltransferase ZDHHC17 and ATG8/LC3 proteins to facilitate golgiphagy (6).
