    Background

    Exosome component 2 (EXOSC2), also known as RRP4, is part of the exosome complex involved in RNA processing and mRNA decay by utilizing 3’-5’ exoribonuclease activity (1,2). The exosome consists of an inert ring of nine subunits coupled with the enzyme DIS3 or DIS3L (3-5). EXOSC2, along with EXOSC1 and EXOSC3, forms the cap of the complex and assists in 5.8S rRNA generation and mRNA degradation (1,6). Missense mutations in EXOSC2 alter exosome binding to certain mRNAs and cause a syndrome termed SHRF (short stature, hearing loss, retinitis pigmentosa, and distinctive facies) (7,8). Overexpression of EXOSC2 was shown to promote metastatic breast cancer in a tRNA-dependent manner (9).
