    Background

    Repulsive guidance molecule b (RGMb), also known as DRAGON, acts as a bone morphogenetic protein (BMP) co-receptor and glycosylphosphatidylinositol (GPI)-anchored cell-surface protein belonging to the RGM family. RGMb is composed of an N-terminal signal peptide, a partial von Willebrand factor D-type domain, a hydrophobic domain, and a C-terminal GPI anchoring domain (1). RGMb is expressed on a variety of cell types, including epithelial cells, neuronal cells, and immune cells (1-3). RGMb was first characterized in a study of factors regulated by dorsal root ganglion 11 (DRG11), playing a regulatory role in various processes, including embryonic development, immune function, injury repair, and tumorigenesis (2-8). RGMb enhances BMP signaling at the cell membrane and can regulate cytoskeletal rearrangement via the neogenin-Rho pathway regulation (9,10). RGMb has also been shown to interact with PD-L2, and the downregulation of the PD-L2/RGMb pathway is a mechanism by which the gut microbiome can promote responsiveness to PD-1 immune checkpoint blockade (ICB) (11).
