    Background

    U6 biogenesis 1 (USB1), also known as MPN1, is a 3’-5’ RNA exonuclease that removes uridines and adenosines from the 3’ end of U6 snRNA, creating a 2’, 3’-cyclic phosphate formation, which protects U6 snRNA from degradation by the nuclear exosome (1-3). USB1 measures the U6 oligo(U) tail by reading the A102 nucleotide position and pauses five uridine residues downstream (3). Loss of USB1 results in a rare condition called poikiloderma with neutropenia, where U6 snRNA is unstable and contains aberrantly long and oligoadenylated 3’ ends, resulting in splicing dysfunction (1,4,5).
