    Background

    Ubiquitination is a critical posttranslational modification (PTM) that regulates numerous proteins in eukaryotic cells via the ubiquitin-proteasome system (UPS). Homologous to the E6AP carboxyl terminus (HECT) proteins are a family of E3 ubiquitin ligases that catalyze the transfer of ubiquitin to target proteins, marking them for proteasomal degradation (1). HECT proteins can be further classified as HECT and RCC1-like domain-containing (HERC) proteins, which possess a catalytic HECT domain and one or more RCC-like domains (RLDs) (2). HERC4, a member of the HERC family, is overexpressed in various cancers, such as hepatocellular carcinoma and breast cancer (3-6). HERC4 is known to target several proteins for ubiquitination, including progesterone receptor (PGR), c-Maf, Smoothened (Smo), salvador homolog (SAV1), and keratin 19 (K19) (4,7-11). HERC4 can also mediate the clearance of proteins that have been targeted with molecular glue degraders (MGDs) or proteolysis-targeting chimeras (PROTACs). For instance, the mechanism of action of a small-molecule degrader of STING was found to be mediated in part by HERC4 (12).
