    Background

    DNA replication stress, a major source of genome instability, results from barriers to DNA replication fork progression, including DNA damage lesions, transcription-replication conflicts, and nucleotide depletion. Multiple pathways are in place to stabilize replication forks under stress, and to promote DNA replication restarting (1). PrimPol is a nuclear and mitochondrial primase and DNA-directed polymerase belonging to the archaea–eukaryotic primases (AEP) superfamily. PrimPol catalyzes prepriming of DNA after replication stalling, allowing DNA replication to continue in the presence of DNA damage. The repriming occurs beyond the DNA lesion, leaving small single-stranded DNA (ssDNA) gaps that are subsequently repaired (2,3). PrimPol abundance is regulated by ubiquitin-mediated proteasomal degradation, and deubiquitination by USP36 stabilizes PrimPol in response to replication stress (4). PrimPol activity is negatively regulated through phosphorylation by PLK1 at Ser538 (5), and positively regulated through ATR/Chk1-dependent phosphorylation at Ser255 (6). Aberrant expression of PrimPol has been observed in human cancers, and its expression in cancer may confer vulnerability to therapies targeting DNA repair pathways (2,7).
