    Background

    Macrophage receptor with collagenous structure (MARCO) is a class-A scavenger receptor (SR) type II transmembrane glycoprotein and part of the broader family of pattern recognition receptors (PRRs) (1,2). MARCO is expressed on alveolar macrophages and Kupffer cells in the liver, and macrophages in other tissues, such as lymph nodes, spleen, and intestines (3,4). MARCO plays an important role in host defense by sequestering potentially pathogenic viruses and bacteria, and it mediates the binding of unopsonized particles such as TiO2, Fe2O3, silica, and carbon nanotubes (3,5-7). In recent studies across multiple cancer types, increased infiltration of MARCO+ tumor-associated macrophages (TAMs) correlated with poor patient prognosis (8). Due to its role in regulating myeloid-derived suppressor cell (MDSC)-mediated immunosuppression, MARCO has been identified as a therapeutic target for cancer (9).
