    Background

    DOCK8 (dedicator of cytokinesis 8) is a member of the DOCK family of proteins that function as guanine nucleotide exchange factors (GEFs) for the Rho family of GTPases (1,2). DOCK8 is responsible for activation of the GTPase Cdc42, which exchanges bound GDP for free GTP, and plays a regulatory role in cell polarity and migration (2). DOCK8 immunodeficiency syndrome, or autosomal-recessive hyper-IgE syndrome, is caused by mutations of DOCK8 (3,4). Patients with this disorder have susceptibility to viral infections, eczema, increased serum IgE, defective T cell activation and differentiation, and impaired eosinophil homeostasis, due to T cell dysfunction associated with DOCK8 deficiency (2-4).
